The suppression of AD-3B mutants by supersuppressors in Neurospora crassa.

چکیده
The action of eight suppersuppressors has been tested on 76 ad-3B mutants which were induced by base-pair transition mutagens. Thirteen mutants were found to be suppressible by at least one ssu gene. Most of the suppressible mutants were found to belong to a class in which one would expect to find nonsense mutants (polar complementing or noncomplementing classes, with AT at the mutant site). However, suppression was observed in other classes, including those containing presumed missense mutants. The specificity of the suppressors and the criteria for molecular classification of the mutants are discussed.

Mutation induction by difunctional alkylating agents in Neurospora crassa.
The genetic characterization of ad-3 mutants of Neurospora crassa induced by two carcinogenic difunctional akylating agents, 1,2,4,5-diepoxypentane (DEP) and 1,2,7,8-diepoxyoctane (DEO), has shown that point mutations at the ad-3B locus have similar complementation patterns. In addition to the induction of point mutations, DEP induces a low frequency (7.5%) of multilocus deletions, whereas DEO ...

conditions was the compound found to be mutagenic. Mutants were induced in a gentically marked 2-component heterokaryon and recovered as purple colonies among the white, nonmutant background colonies. In this system, purple colonies result from two types of genetic damage in the adenine-3 region: point mutations at ad-3A and ad-3B loci and multilocus deletions covering one or both loci simultan...
متن کاملBiotransformation of Hydrocortisone by Neurospora crassa
The ability of Neurospora crassa FGSC 4335 in the biotransformation of hydrocortisone was investigated. The microorganism produced two major metabolites after incubation with the substrate for seven days. Each microbial product was purified chromatographically and identified on the basis of spectral data. The products were identified as 11?,17?,20?,21-tetrahydroxypregn-4-en-3-one (II) and 11?-h...
متن کاملEnzymology of the pigmented adenine-requiring mutants of Saccharomyces and Schizosaccharomyces.
The red or purple adenine-requiring mutants of Saccharomyces, Schizosaccharomyces, and Neurospora have been categorized genetically as resulting from the mutation of two separate loci. In Neurospora crassa these are the ad-3A and ad-3B loci (de Serres, 1956); in Saccharomyces cerevisiae, the ad-l and ad-2 loci (Roman, 1956); and in Schizosaccharomyces pombe, the ad-6 and ad-7 loci (Leupold & Gu...
متن کاملAn Enzymatic Difference among Pyr-3 Mutants of Neurospora Crassa.
The pyrimidine-3 mutants of Neurospora crassa have received considerable attention in the past. Mitchell and Mitchell' described experiments, using pyr-3a (37301), 3b (37815; temperature sensitive), and 3d (45502), which indicated that the pyr-3a and 3b mutations represent a genetic unit distinct from 3d. The evidence for this view was that pyrimidine-independent heterocaryons could be formed b...
